Reduction of Wrinkles and Fine Lines and Increased Skin Resiliencey |

| The previously described scientific data showed increased levels of newly synthesized tropoelastin in cell culture and increased cellular density in human skin (punch biopsies) as direct effects of HEP. A ramification of these data is that use of HEP, applied twice daily, effectively lessens the appearance of wrinkles and fine lines. Moreover, topically applied HEP returns elasticity and resilience to skin as shown through cutometer measurements. |
| The cutometer measures skin resilience, an element of which is elasticity. After 8 weeks of twice-daily use, the average improvement of skin resilience was approximately 145%. | A unique property of elastin is the hormonally active peptides hidden in its structure. These are released through the action of certain enzymes, either produced by the cells of the matrix or supplied from an external source as in a laboratory environment. They form the active ingredients of the proprietary peptide mix contained in Elastica which we manufacture under extremely carefully controlled conditions using an enzyme called thermolysin. There are at least a dozen of these bioactive peptide sequences within the structure of elastin. They are small, each less than 1000 Daltons. They consist of such amino acid strings as valine-valine-proline-glutamine (VVPQ) or valine-alanine-proline-glycine (VAPG). What seems to happen in our bodies is that matrix cells release enzymes called proteases during a remodeling event. Proteases break down the matrix proteins to remove them to make room for new proteins to be laid down. The action of the proteases releases the bioactive peptides of the elastin. These attach to specific receptors on the matrix cell surfaces. These attachments start a cascade of events going on inside the cells resulting in cell division and or the turning on of protein export machinery to put new collagens and elastin into the extracellular matrix. The end result is a restructured matrix with many more cells as well as enhanced extra-cellular components in the form of collagen and elastic fibers. |

| Elastin is a rubber-like protein found in virtually alll tissues of the body. Elastin lends elastic recoil to tissues of which it is a significant component, eg. skin, lung, aorta. Elastin's secondary function contains peptide sequences with hormonal activity which is important in tissue remodeling. |
| Elastin is a highly cross-linked complex polypeptide and is a major component of elastic fibers present in the skin and connective tissue of animals. Elastin appears to be primarily responsible for the physiological elasticity of tissue. In normal mammalian skin, specifically human skin, elastic tissue proteins represent a relatively small fraction of the total dermal proteins, but play a very important role in maintaining or improving the skin tone and structure. Elastin itself is the main protein substance present in elastic fibers and occurs in tendons, blood vessels, and connective tissue. When isolated from these sources, it is normally in the form of a brittle. fibrous, yellowish material which is insoluble in water, alcohol and ether but is somewhat soluble in concentrated aqueous alkali metal hydroxide solutions. The dense cross-linked structure of elastin makes it very difficult to solubilize. There have been many attempts to solubilize elastin and create cosmetic agents from the solubilized elastin. To date, these techniques have not found a large degree of commercial success. |
| Attempts to solubilize have been attempted, however, it has been found that elastin is only slightly absorbed by the skin and does not sufficiently penetrate the skin to produce substantial benefits to the skin. UNTIL NOW! |

| Discovered in the 1980's when Drs Sandberg and Roos were doing metabolic studies on elastic tissue turnover. HEP, which stands for Hydrophilic (water-loving) Elastin Peptides is produced by thermolysin (an enzyme) digestion of highly purified elastin and is made up of 44 small discrete peptides. |
| HEP is directed to an elastin peptide fragment which is useful as a therapeutic and or cosmetic composition or agent for modifying tissue, especially skin. The term "modify" is used to convey that the present invention changes either the appearance, form, characteristics and/or the physical attributes of the tissue to which it is being provided, applied or administered. The change in form can be reflected in any of the following alone or in combination: enhanced appearance of the skin; increased softness of the skin; increased turgor of the skin; increased texture of the skin; increased elasticity of the skin; decreased wrinkle formation and increased endogenous elastin production in the skin. |
| This invention relates generally to a composition useful in treating tissue and a method for treating tissue, wherein the composition utilized to treat the tissue includes a peptide which simulates the effect of elastin, and more preferably increases the native production of elastin. Preferably the peptide has an amino acid sequence which corresponds to or is homologous with a fragment of mammalian elastin, more preferably the amino acid sequence corresponds or is homologous with fragments of elastin endogenous to the tissue being treated. It is preferable that the peptide is at a therapeutically effective concentration and/or is an active ingredient of a therapeutic or cosmetic composition. More specifically, the present invention relates to a composition which includes a peptide or multiple peptides which increase the elasticity and turgor of the skin. The present invention is also directed to a method of administering the composition to a patient in need thereof to thereby increase the elasticity of the patient's skin. Increased elasticity results in an improvement in the overall physical appearance of the skin. For example, when treated with the present invention, mammalian skin appears to be softer, smoother, healthier, and more youthful. |

| Skin conditions: There are many skin conditions and diseases which would benefit from elastin treatment. Beyond the obvious cosmetic applications (i.e., increased tone, turgor, and appearance), enhanced elastin production will produce long-term beneficial results. For example, the inherited disease Scleraderma is characterized by a thickening and stiffening of the skin, and cutaneous ulcers due to the overproduction of collagen (there are a number of diseases which involve overproduction of collagen and which seem to have an adverse effect on elastin production/content and compromise the tissue). This disease can also have systemic effects on organs and blood vessels. The stiffness and difficulty in motion along with the cutaneous ulcers would benefit greatly from incorporation of elastin into the skin. A similarly positive outcome would be expected with the treatment of lupus and rheumatoid related skin changes which are generally collagen-vascular diseases involving a decrease in elastin. |
| Other skin conditions would appear to benefit from the present invention. Conditions and problems such as acne rosacea, acne vulgaris, aging skin with vascular fragility, burn treatment, scar contractures from burns, radiation burns, pruritis (or chronic itching), psoriasis, urticaria (commonly referred to as hives), xerosis (abnormal dryness of the skin, eyes or mouth), vesicular dermatoses, cracked fingers and feet, drug eruptions (from an allergic reaction), epidermolysis (a skin condition where the epidermis is in a loosened state, often with the formation of blebs and bullae either spontaneously or after trauma), and erythema multiforme would benefit from treatment with the elastin peptide(s) of the present invention. |
| Hair: Hair growth, color, and removal can all be improved by treatment with elastin peptides which will make the hair stronger, more shiny, and improve the condition and healing of irritated skin upon removal of unwanted hair. Premature graying of hair may also be due to decreased elastin. |
| Lips: Chapped lips and chronic dermatitis or inflammation of the lips can be greatly improved upon treatment with elastin peptides of the present invention. Long-term relief would be a potential benefit from the stimulation of endogenous elastin in these tissues. |
